چکیده

The appearance of P-adrenergic receptors and isoproterenol-stimulated adenylate cyclase during differentiation of embryonic quail muscle cells in tissue culture has been examined. P-Adrenergic receptors first appear during the fusion stage of myogenesis. ['"I]]Iodohydroxybenzylpindolol (['2sI]IHYP) was used as the ligand to characterize &adrenergic receptors. In equilibrium binding studies, mature myotubes contained, per diploid nucleus, approximately 800 specific and sa turable binding sites, which have a dissociation constant ( K d ) for ['251]IHYP of 108 PM. When determined from the association and dissociation rate constants, the kinetically derived Kd is 63 PM. ['2sI]IHYP binding to myotube membranes is blocked stereoselectively by a number of P-adrenergic agonists and antagonists. In contrast, proliferating myoblasts contain fewer than 10 /?-adrenergic receptors per cell. Both myoblast and myotube membranes contain an adenylate cyclase activity which is stimulated by guanine nucleotides and fluoride, and is activated in membranes derived f rom cells treated with cholera toxin. Myotube membrane adenylate cyclase, but not myoblast membrane adenylate cyclase, is stimulated 2to %fold by isoproterenol. Accumulation of CAMP by intact myotubes, but not by myoblasts, is stimulated 3to &fold by isoproterenol. Both cell types accumulate CAMP in response to cholera toxin. These results indicated that as the embryonic muscle cells fuse, they develop specific P-adrenergic receptors that become functionally coupled to a pre-existing adenylate cyclase.

Expression of genes for metabolism of cyclic adenosine 3':5'-monophosphate in somatic cells. beta-Adrenergic and PGE1 receptors in parental and hybrid cells.

Using the ligands [125I]iodohydroxybenzylpindolol and [3H]prostaglandin E1 ([3H]PGE1), we have studied the relationship of receptors for beta-adrenergic agents and for PGE1 to adenylate cyclase in membranes of parental, hybrid, and variant mammalian cell lines. Fusion of parental clones responsive to beta-adrenergic agonists (beta+) with unresponsive clones (beta-) produced hybrid clones with a...

Internalized Receptors Reconstitute Catecholamine-stimulated Adenylate Cyclase Activity*

The adenylate cyclase-coupled &-adrenergic receptor of the frog erythrocyte has served as a useful model system for elucidating the mechanisms of catecholamine-induced desensitization. In this system, it has been previously demonstrated that agonist-induced refractoriness is associated with sequestration of the @adrenergic receptors in vesicles away from the cell surface and from their effector...

Catecholamine-induced desensitization of turkey erythrocyte adenylate cyclase is associated with phosphorylation of the beta-adrenergic receptor.

Preincubation of turkey erythrocytes with catecholamines desensitizes the beta-adrenergic receptor-adenylate cyclase complex in the plasma membranes of these cells. Photoaffinity labeling of the beta-adrenergic receptors with 125I-labeled p-azidobenzylcarazolol (125I-pABC) and subsequent analysis by NaDodSO4/polyacrylamide gel electrophoresis demonstrates an altered mobility of receptor peptide...

Anti-alprenolol anti-idiotypic antibodies bind to beta-adrenergic receptors and modulate catecholamine-sensitive adenylate cyclase.

Rabbit antibodies induced against alprenolol, a potent beta-adrenergic antagonist, bind to other antagonists, and, with less avidity, to catecholamine agonists. Anti-idiotypic antibodies were raised against the anti-alprenolol immunoglobulins. The anti-idiotypic antibodies specifically bind and agglutinate turkey erythrocytes; this is not observed for human agglutinate turkey erythrocytes; this...
